The incident happened at the Co-Op store in Filey Close at around 8.50pm on Thursday, February 14.
It was reported that a man had approached the cashier and told her to put two bottles of wine and a quantity of cash in a carrier bag.
He then threatened her before leaving the store with a second man who was waiting by the doorway.
Both suspects made off on foot in the direction of Scarborough Avenue.
Detective Constable Neil Davies, from the Stevenage Local Crime Unit, said: “The cashier was not injured during the robbery but she has been left understandably shaken by the ordeal.
“I am appealing to anyone who recognises these two men to please come forward and speak to us, as I believe they have information that could assist us with our enquiries.
“I am also keen to hear from people who saw anything suspicious in the area around the time of the incident. “I’d like to take this opportunity to reassure residents that this type of crime is unusual in Stevenage, and we are doing all we can to trace those involved.”
